Even an initial submission must follow the standard formatting requirements unless others are stated.
Double spaced, 12pt Times New Roman (or Courier), 1 inch margins all round.
Number your pages in the bottom right hand side of the footer. Include your name and the title of the work elsewhere (I put it at the bottom left hand side of the footer but I don't know of the "official" guidelines on that one) in case it's printed and any of the pages get separated. I usually also include a cover sheet with my name and contact details on it.
That said, ALWAYS double check the guidelines of the agency/publisher. Unless otherwise stated, you should be fine with the standard.

You ought to try checking if the agent has specific requirements first. Many have a website with this sort of info.
IME most initial submissions need 3 chapters (or a word limit) with a synopsis and covering letter. It's often only full (hard copy) submissions where they'll specify font, spacing and margins, though it makes sense to use a standard-ish font in 11 or 12-point to make contact, so it's easy to read.
