Prologue and First Three Chapters

When an agent asks for the first three chapters, do you include the prologue and then chapters one-three or do you include the prologue and chapters one-two?

I think I'm revising the first three chapters for the last time before I start sending them out again so wish me luck, however chances are I will have to start revising them again after I've sent them to just the one agent!
